Three teams of Instytut Genetyki Człowieka PAN conducting research on hematologic malignancies actively implemented and through participation in the project have improved their scientific performance.

🩸A team from the Department of Molecular and Clinical Genetics led by Prof. Małgorzata Dawidowska has a long-standing collaboration with Universiteit Gent, which was invoved in project.

Prof. Dawidowska delivered within the project a lecture entitled ‘Multi-omics analysis in T-cell acute lymphoblastic leukemia: in search for biological mechanisms and clinical meaning’.

The team organized 2021 dedicated to ‘Acute Lymphoblastic Leukemia’ and also diseeminate research results on international conferences such as ‘3rd Translational Research Conference: Acute Lymphoblastic Leukaemia’ and ‘European Hematology Association (EHA) 2023 Congress’.

As a result of the project's collaboration with partners from Ghent was published joint research paper entitled ‘Multiomics to investigate the mechanisms contributing to repression of PTPRC and SOCS2 in pediatric T-ALL: focus on miR-363-3p and promoter methylation’.

The leukemia team is implementing in the international cooperation 3 hemato-oncological projects:
▪️‘Identification of mTOR pathway deregulation mechanisms alternative to mutations in T-cell acute lymphoblastic leukemia (T-ALL)’
▪️‘Identification of genes and mechanisms underlying the recurrence of T-cell acute lymphoblastic leukemia’
▪️‘CircRNAs involved in mechanisms of malignant transformation and relapse in T-ALL: towards new RNA-based therapies’
and is waiting for evaluation results of another one within programme.

🧬The Molecular Pathology Department team is led by two P*s: prof. Agnieszka Dzikiewicz – Krawczyk and prof. Natalia Rozwadowska. The team conducts research focusing on the genetics and molecular biology of hematologic malignancies (leukemia, B-cell, and T-cell ).

The team works extensively with the prof. Anke van den Berg's group of UMCG, which invited to participate in the project consortium.

Young researchers from this team have participated to the greatest extent in the mobility offered by the project.

Prof. Dzikiewicz – Krawczyk delivered within the project a lecture ‘Mortal Kombat: fighting MYC in hematologic malignancies’

The team prepared workshop ‘Advanced imaging of biological samples’ and Hematology Day 2022 ‘CAR-T therapy in hematologic neoplasms’ and took part in many networking activities to strengthen international cooperation.

In addition, the researchers participated in conferences such as '4th International Conference on the Long and the Short of Non-Coding RNAs'n and EMBO Workshop ‘Non-coding RNA medicine’.

The project collaboration has resulted in two joint publications:
▪️ 'CRISPR/Cas9 screen for genome-wide interrogation of essential MYC-bound E-boxes in cancer cells'
▪️ 'Inhibition of the glutamate-cysteine ligase catalytic subunit with buthionine sulfoximine enhances the cytotoxic effect of doxorubicin and cyclophosphamide in Burkitt lymphoma cells '
and implementation of hemato-oncological project entitled ’Overcoming CAR-T cell exhaustion by modifying enhancers using CRISPR technology’.

The team submitted 3 projects under , including , which is in the process of evaluation.

🧪Prof. Maciej Giefing is leader of Department of Cancer Genetics team, which focuses on genomic and epigenomic alterations that contribute to the development of neoplasms, especially classic .

The team is collaborating with Prof. Reiner Siebert's research group at , which was invited to participate in the project.

Prof. Giefing delivered within the project a lecture entitled ‘Unusual expression pattern of the negative NF-ĸB regulator miR-181a and its role in classical Hodgkin lymphoma’.

His team organized workshop ‘DNA bisulfite pyrosequencing – from whole genome to targeted gene methylation analysis' and Hamtology Day 2023 ‘Hodgkin Lymphoma – the great teacher’.

The team disseminated research on such conferences as ‘EMBL Conference Chromatin and epigenetics’ and ‘17th International Conference on Malignant Lymphoma’.

The researchers have published joint paper entitled ‘Identification of two unannotated miRNAs in classic Hodgkin lymphoma cell lines’ and implement hemato-oncology research projects in cooperation with foreign partners:

▪️ 'cfDNA methylome analysis of patients with Hodgkin's lymphoma'
▪️ 'BCOR alterations and the activation of CXCL12 oncogenic pathway in classical Hodgkin lymphoma'
▪️ 'Functional analysis of novel oncomiR candidates in classical Hodgkin lymphoma'.

Recently, the international cooperation with German partners resulted in publication entitled ‘Transcriptional reprogramming by mutated IRF4 in lymphoma’ published in Communication.

Twinning projects step up scientific excellence of research organisations in countries. We present the impact of project implemented by consortium cosisting Instytut Genetyki Człowieka PAN, UMCG - Universitair Medisch Centrum Groningen, Universiteit Gent and Universität Ulm.

🧬 Project became a challenge due pandemic which started before we launched the project. 💻Most of the meetings related to project management had to take place online. Main communication tool was email and online platform.
Despite these difficulties, we were also able to implement the project thanks to one very important thing - trust and cooperation. Project was implemented by partners, who already knew each other well and worked together. This ensured that we had good contact, understanding and respect within the consortium.

👩‍🦳👱👨‍🦰👩The team involved in the project consisted of experiences of all the partners: Prof. Maciej Giefing (project coordinator) from the Instytut Genetyki Człowieka PAN, Prof. Anke van den Berg from University Medical Center Groningen, Prof. Pieter Van Vlierberghe from Ghent University and later Prof. Panos Ntziachristos and Prof. Reiner Siebert from Ulm University.
Excellent P*s: Prof. Natalia Rozwadowska, Prof. Małgorzata Dawidowska and prof. Agnieszka Dzikiewicz – Krawczyk supported the strategic management of the project and were involved in many project activities. The project manager was Agnieszka Możejko.

😞There was a difficult time when we lost Professor Pieter Van Vlierberghe. However, we appreciate the knowledge he passed on to the project participants and his great contribution to the field of hamatoncology, which we still benefit from.

🥰Special thanks to people who supported us in project writing and implementation:
Anna Stachowiak-Szrejbrowska, who supported us in writing the proposal and provided a lot of practical advice related to project implementation and evaluation, and two experts of Polska Zachodnia:

Daniel Sikora from Instytut Genetyki Człowieka PAN is finishing his training stay within project.
🩸He have been visiting Professor Reiner Siebert’s team at Universität Ulm. The group is very experienced in the molecular biology research and is well recognised in the field, so Daniel had a chance to get to know state of the art and technology regarding epigenetic studies.

💻Additionally, thanks to the strong bioinformatical base of the team, he was introduced to the pipeline of the data analysis obtained via various methods, what will be later useful in processing and visualisation of his own data.

🧬Daniel’s Ph.D thesis is “cfDNA methylome analysis in Hodgkin lymphoma patients” and the experience and knowledge obtained on this traineeship will be useful in analysing the methylation of lymphoma as well as other hematologic cancers.

🧪Work with the internationally recognised researchers is an excellent opportunity to broaden horizons, to learn new approaches to lymphoma research but also about other diseases.

and outreach activities implemented within the project aimed to increase the visibility and recognition of the Instytut Genetyki Człowieka PAN as well as the consortium partners: Universiteit Gent, UMCG - Universitair Medisch Centrum Groningen and Universität Ulm.

🩸🩸🩸We organized 3️⃣ events to raise awareness of the general public about and , improve knowledge regarding haematological neoplasm among different groups of audience: school pupils, students, patients, physicians, medical and biological students, researchers and specialists.

🔬 Every year we offered different topic related to hemato-oncology malignancies, recent research results and innovative treatments:
• Acute Lymphoblastic
• -T Therapy in Hematologic Neoplasms
• Lymphoma – the Great Teacher

Lecturers at these events were prominent Polish and foreign specialists: Prof. Katarzyna Derwich and Lidia Gil from Uniwersytet Medyczny im. Karola Marcinkowskiego w Poznaniu, Prof. Tomasz Szczepański from Śląski Uniwersytet Medyczny, Prof. Jan Maciej Zaucha from Gdański Uniwersytet Medyczny, Prof. Ralf Küppers from Uni DuE, Prof. Grzegorz Basak from University Clinical Center of Medical University of Warsaw.

🧪In addition to experienced specialists, activities for the youngest participants of Hematology Day were led by young scientists of IHG PAS: Monika Drobna, Marta Kasprzyk, Greta Sawicz and the institute's popularization team.
Hematology days gathered over 6️⃣0️⃣0️⃣ participants.

🧬The popularization team of the IHG PAS organized also the as part of the organized on the occasion of the 25th anniversary of the Wielkopolska Region. Our specialists offered for the wide audience workshops and games, which learned about the basics of genetics, hematological diseases, the importance of in the functioning of the body and in the formation of lymphocytes. The event was attended by nearly 🔟0️⃣0️⃣0️⃣ people.

👨‍🏫Researchers involved in the project disseminate haematological research results on over 1️⃣6️⃣ conferences, symposia and seminars.

The project results were disseminate during webinar 'How to strategically use funds under the Widening Horizon Europe program', which was organized by HPK Polska Zachodnia and the Biuro Wielkopolski w Brukseli.

Creating strong international partnerships was implemented within project through networking activities.

🔹Staff involved in project implementation took part in brokerage events, seminars, conferences and info days, such as 'Cluster Health Brokerage and Info Days', 'Central European BioForum' etc.

🔹To improve organizational capacity and introduce changes leading to effective research management at the Genetyki Człowieka PAN 4️⃣ Best practice exchanges - short stays of Polish P*s at Universiteit Gent , UMCG - Universitair Medisch Centrum Groningen and Universität Ulm took place.

🔹In addition, 4️⃣ P*s from IHG PAS took part in Management related training organized by Ghent University to increase chances of successful obtaining competitive funds for research and innovation. It also allowed to improve skills in managing research groups, time and risks.

🔹2️⃣ ESRs meetings dedicated to professional career development were organized, during which young researchers developed professional skills crucial for their scientific careers in the area of scientific communication and scientist’s personal branding on social media. They learned how to navigate the grant writing process, how to prepare a successful application, and how to win a grant. The events supported also participants’ soft skills related to stress handling and asking for help.

Participation in networking activities supported the development of scientific contacts and the building of collaborations for joint research projects😀

We strengthened knowledge and innovation transfer through✈️international mobility within WP3 Stepping up research excellence. At the beginning we struggled with the difficulties caused by the 🦠COVID- 19 pandemic. 😀Fortunately, our participants were fearless and the institutions implementing the project were professional, so in the end we were able to carry out the planned activities.

As part of the project, we implemented 1️⃣2️⃣ study visits for scientists at every stage of their careers and 8️⃣ three-months training stays of doctoral students from the Instytut Genetyki Człowieka PAN to partner universities.
Ph.D. students from Universiteit Gent, Universität Ulm and UMCG - Universitair Medisch Centrum Groningen also had the opportunity to visit the IHG PAS in the framework of week-long visits.

🔬Mobility carried out within the framework of the project has brought significant results. It made possible to agree on new joint research topics, which are being developed in bilateral and multilateral cooperation and has fostered the creation of new project ideas.
Strengthening cooperation has brought a significant impact on IHG PAS. We are pleased with the increase in activity in applying for international projects. Recently, the institute has submitted 8️⃣ proposals (5 for Programme).
🩸Hemato-oncology teams have also increased application to Polish national agencies. Young as well as experienced scientists apply for the projects.

In addition, the international mobility implemented under the project had the following impact:
◾️ supported the training and qualification of research staff by increasing their experience working in foreign laboratories and with international teams
◾️ increased the level of interdisciplinarity of research by enabling the development of cooperation involving other disciplines
◾️ enabled the establishment of links between different research groups that will result in joint research and scientific publications in the future.

🔸 ‘Advanced imaging of biological samples’

📑To strengthen other skills and develop awareness of other issues corresponding the conduct of scientific research, 4️⃣ Soft Skills Seminars, open to all, were organized, covering the following topics:
◾️ ‘Experiments with the use of laboratory animals – is it so difficult? Practical, ethical and legal aspects of working with laboratory animals’
◾️ ‘Entrepreneurship’
◾️ ‘IPR in research institutions’
◾️ ‘Storytelling’

🔬The project paid special attention to the development of Early Stage Researchers. In order to straighten their qualifications and profession development 'Transferable skills workshop' was provided.

Almost 3 years ago we have launched ▶️NEXT_LEVEL project. Its main goal is to step up research excellence in the field of 🩸🧬🧪hemato-oncology of Instytut Genetyki Człowieka PAN and cooperating research organisations: UMCG - Universitair Medisch Centrum Groningen, the Netherlands, Universiteit Gent, Belgium, Universität Ulm, Germany.
The implementation of the project has significantly contributed to the scientific development of the participating institutions, especially the IHG PAS>

🇪🇺The project is funded from Twinning Action of Horizon 2020 programme, which is a great mechanism to support research institutions in widening countries such as Poland.

🥳We are happy with the successes of young researchers. Marta Kasprzyk from Instytut Genetyki Człowieka PAN - participant of our project, speaker on Hematology Day 2022, has just defended her Ph.D thesis entitled 'Functional dissection of IGH enhancers and enhancer RNAs in B-cell non-Hodgkin lymphomas'.
