Using our peptide delivery platform, we are advancing a pipeline of novel Precision Endocrine Peptides (PEPs)™ designed to overcome key limitations of existing treatments and improve efficacy, tolerability, safety, and convenience. Our lead candidate, canvuparatide (MBX 2109) – a potential once-weekly parathyroid hormone (PTH) replacement therapy for the treatment of chronic hypoparathyroidism – i

s preparing for Phase 3 development. We are also advancing an obesity portfolio including MBX 4291, a GLP-1/GIP co-agonist prodrug in Phase 1 development, and MBX 5765, an amycretin prodrug in preclinical development. In addition, we are on track to nominate its GLP-1/GIP/GCG receptor prodrug candidate in Q3 2026, further expanding its obesity pipeline to potentially address the full spectrum of patient needs. Today we are excited to announce that the first patient has been dosed in the pivotal Phase 3 oPTimize trial evaluating its once-weekly, investigational parathyroid hormone (PTH) replacement therapy for the treatment of adults with chronic hypoparathyroidism (HP).

The global registrational study will enroll approximately 160 patients and builds on positive Phase 2 Avail™ results and durable one-year data demonstrating sustained clinical benefit.

We look forward to advancing the oPTimize trial as we work to bring this potential therapy one step closer to people living with HP.
