Alera Bio announces the appointment of David McCullough as Chief Executive Officer.

David is an accomplished biotechnology executive with extensive experience in corporate strategy, capital formation, operational ex*****on, and the advancement of emerging therapeutics companies. He has successfully guided biotechnology organizations through critical stages of growth, financing, and strategic development.

As CEO, David will lead Alera Bio’s corporate strategy, financing initiatives, organizational development, and ex*****on of the company’s clinical and regulatory roadmap as we advance AB-101 for MCT-8 deficiency (Allan-Herndon-Dudley Syndrome).

This agreement expands our intellectual property platform supporting the development of therapies for MCT-8 deficiency (Allan-Herndon-Dudley Syndrome).

Dr. Eisen is a seasoned physician-scientist with over 20 years of experience spanning preclinical research through early-stage clinical development. His expertise in neuroscience, metabolic disease, and translational medicine aligns directly with our mission to develop transformative therapies for patients with rare neurological disorders.

In his new role, Dr. Eisen will lead the advancement of AB-101, our novel multi-mechanism therapy for MCT-8 deficiency — a devastating ultra-rare condition with no approved treatments addressing neurological symptoms.

With the passage of the Give Kids a Chance Act, the Rare Pediatric Disease Priority Review Voucher (PRV) program has now been reauthorized, reinforcing a critical incentive for companies advancing therapies for children with serious, rare conditions. It is a win for the MCT-8 families who have waited far too long for progress.

In 2024, Alera’s lead program, AB-101 for MCT-8 deficiency, received Rare Pediatric Disease Designation (RPDD) from the FDA. This designation, now paired with a reauthorized PRV pathway, represents more than a regulatory milestone. It validates the urgency of this disease, strengthens the development pathway, and reinforces our commitment to advancing a therapy for children who currently have no approved treatment options.

We are grateful to the advocates, clinicians, policymakers, and researchers who made this reauthorization possible, and we remain focused on translating this momentum into meaningful outcomes for patients.

Alera Bio has entered an exclusive licensing agreement with BLA Technology, LLC to advance AB-101, our lead program for MCT-8 deficiency (Allan-Herndon-Dudley Syndrome).

MCT-8 deficiency is a devastating, ultra-rare neurological disorder that affects ~1 in 70,000 males worldwide. It leads to profound neurodevelopmental delays, severe motor dysfunction, cognitive
impairment, and systemic thyroid complications. Today, there are no approved therapies.

AB-101 has already received FDA Rare Pediatric Disease designation and is being advanced as a program designed to address both neurological and endocrinological symptoms — a dual-impact approach that could transform care for patients and families.
