A recent review published in Antibodies explores the multiple mechanisms through which anti-TNF-α antibodies may help control inflammation in inflammatory bowel disease (IBD), including Crohn’s disease and ulcerative colitis.
While anti-TNF-α therapies are well known for neutralizing soluble TNF-α, the review highlights additional actions involving membrane-bound TNF-α, including:
• Complement-dependent cytotoxicity
• Antibody-dependent cellular cytotoxicity
• Apoptosis of membrane TNF-α–expressing cells
• Apoptosis of lamina propria T cells
• Induction of regulatory T cells and macrophages
These multifaceted mechanisms may help explain the effectiveness of anti-TNF-α antibodies such as infliximab, adalimumab, and golimumab in reducing IBD-associated inflammation.
Understanding how therapeutic antibodies act beyond simple cytokine neutralization continues to provide valuable insight into antibody-based therapies and inflammatory disease research.
