This , we’re highlighting how mRNA technology could unlock cancer targets researchers have known about for decades but haven’t been able to drug.

Over the last few decades, cancer researchers have identified multiple biological pathways that could be harnessed to help the immune system attack tumours. Yet figuring out how to activate these pathways has proved challenging.

At ME Therapeutics, we’re applying mRNA technology and targeted delivery to one such target: the STING (Stimulator of Interferon Genes) pathway. Our recent preclinical data demonstrates the ability to more selectively activate STING in tumours, providing early evidence for this approach.

In an interview with Onyx Newsroom, CEO Salim Dhanji shares how we were founded to solve the challenge of overcoming immune suppression in the tumour microenvironment to allow a patient's own immune system to fight their cancer.

In a wide ranging discussion, Salim discusses:
- activating the STING inflammation pathway in cold tumours
- moving from ex vivo CAR manufacturing to in vivo CAR manufacturing via a simple infusion
- the experience of being a small, nimble company in the fast-moving mRNA therapeutics space.

We’ve released an update on our in vivo CAR and therapeutic mRNA programs:

▫️ Our in vivo CAR program successfully yielded two lead constructs, each engineered with a distinct approach to targeting both CD19 and CD22 to treat certain blood cancers and autoimmune diseases. Head-to-head preclinical testing is now underway to inform the selection of a final candidate.

▫️ Our therapeutic mRNA program has developed a novel liver-detargeted formulation designed to focus STING expression within the tumour microenvironment. In vitro testing demonstrated significantly decreased STING protein expression in a human hepatocyte cell line whereas expression in non-hepatocyte cells remained relatively unchanged.

▫️ On the corporate front, we continue to move forward on our pathway toward a proposed NASDAQ listing.

In part two of our summer series, we explore how in vivo CAR therapy addresses key limitations of traditional CAR therapies. ME Therapeutics is using in vivo CAR to make a highly effective approach to cancer treatment faster, less costly and more available.

Next month: meet our lead candidate.

In a guest article for Business Insight Journal, our CEO Salim Dhanji explores the potential of in vivo CAR therapy to transform cancer treatment by reprogramming a patient’s immune cells inside the body rather than in a laboratory.

He also shares how ME Therapeutics is advancing an in vivo CAR candidate targeting CD19 and CD22, with the potential to expand how and when CAR therapies can be used.

Here’s a closer look at our therapeutic mRNA candidate targeting the STING pathway. 🔽🔽

Tumors often hide from the immune system when STING is not sufficiently activated. Turning on STING is like flicking a switch to make “cold” tumors visible or “hot”.

Key findings from a colorectal cancer model:

– Our candidate led to the specific recruitment of immune cells into the tumor and significantly reduced tumor growth.

– A single dose synergized with a PD-1 checkpoint inhibitor that was ineffective on its own. This could mean our STING candidate may unlock the efficacy of checkpoint inhibitors in otherwise unresponsive tumours.

– Targeted delivery into myeloid cells in the tumor microenvironment may solve past roadblocks observed with drugs targeting STING, reducing systemic side effects.

Coming to you from in San Diego, where we are feeling the momentum in the immuno-oncology space. It’s been a fantastic event so far, connecting with investors to discuss our differentiated science both in our in vivo CAR program and therapeutic mRNA program, along with our positive preclinical data.

If you are here at BIO and haven't connected with us yet, email [email protected].

We are attending in San Diego and looking forward to meeting with investors.

Connect with us to learn more about our differentiated science that overcomes challenges in immuno-oncology, including the suppressive microenvironment of "cold" tumors and the lack of immune recognition of cancer.

- Our in vivo CAR program targets both CD19 and CD22 markers. Both assets that form the lead therapeutic candidate are currently, or have been, derisked in the clinic. We are also optimizing for both T cells and macrophages.

- Our therapeutic mRNA program activates the highly-validated STING immune pathway in the tumor microenvironment. Positive preclinical data has demonstrated a single dose of our lead candidate synergizes with a PD-1 checkpoint inhibitor otherwise ineffective on its own.
