Research Spotlight | SCIENTZ-150 in Ferritin Nanocage Research

How can ferritin nanocages help enable earlier tumor diagnosis and treatment?

A study published in Science China Life Sciences investigated engineered human heavy-chain ferritin (HFn) nanocages for early tumor theranostics. The researchers developed IRDye800-M-HFn nanoprobes for multimodal tumor imaging and an HFn-Dox system for targeted drug delivery.

Study highlight:

“Engineered nanocarriers have been widely developed for tumor theranostics. However, the delivery of imaging probes or therapeutic drugs to the tumor pre-formation site for early and accurate detection and therapy remains a major challenge.”

The study found that HFn could pe*****te pre-solid tumor sites through inflammation-mediated blood vessel leakage and subsequently accumulate in tumor cells through its targeting interaction with transferrin receptor 1. The HFn-Dox system was then used to deliver doxorubicin to early-stage tumor sites.

Importantly, during HFn preparation, the researchers used a SCIENTZ-150 high-pressure homogenizer to break the HFn-pET-30a-BL21 E. coli cells at 4°C.

Choosing the right sonotrode can make a big difference in ultrasonic cell disruption. The right choice helps improve disruption efficiency, protect your equipment, and deliver more consistent experimental results.

So, how do you choose the right one?

Here are 3 key factors to consider: material, power, and sample volume.

1️⃣ Consider the Material: Durable and Efficient

The material of the sonotrode directly affects its service life and performance.

Titanium alloy is a preferred choice for demanding applications. Compared with standard materials, titanium alloy sonotrodes provide more stable energy transmission and better durability, making them especially suitable for frequent and high-intensity cell disruption.

This helps maintain stable performance and reduce wear during long-term use.

2️⃣ Match the Power to Your Application

Different experiments require different power levels.

🔹 For small-volume samples of 0.5–5 mL, a Φ2 sonotrode with 20–250 W is suitable for precise sample processing.

🔹 For larger sample volumes of 500–1500 mL, a Φ28 sonotrode with 1000–1800 W is a better choice.

The key is to consider both your sample characteristics and processing volume, then match them with the appropriate sonotrode size and power range.

3️⃣ Match the Sonotrode to Your Sample Volume

Sample volume is another important factor when selecting a sonotrode.

A properly matched sonotrode helps ensure efficient energy transfer and stable disruption performance, whether you are processing a small laboratory sample or working with larger-volume applications.

5️⃣ Sonotrode Installation & Removal Guide

3 simple steps for safe operation!

Step 1 – Power Off
Switch off the equipment and prepare a torque wrench.

Step 2 – Remove
Loosen the fixing clamp counterclockwise and remove the sonotrode with the wrench.

Step 3 – Install
Align the threads, pre-tighten the sonotrode, tighten it to the specified torque, and secure the support bracket.

⚠️ Important Tips:

❌ Never operate while the equipment is powered on.
❌ Never hit or force the sonotrode during installation or removal.
🔧 Check the tightness of the connection every 3 months.

The right sonotrode means better matching, more efficient disruption, and more stable experimental results.
