→ HIF-1α在常氧下半衰期只有几分钟！
✅ 解决方案：三气培养箱1% O₂ / CoCl₂ 100-200μM / DFO 100μM

❌ 坑2：用普通全细胞裂解液
→ HIF-1α是核蛋白，效率极低！
✅ 用核蛋白提取试剂盒

❌ 坑3：裂解太慢
→ 每多一分钟就降解一分
✅ 全程冰上操作+蛋白酶抑制剂

❌ 坑4：上样量不够
✅ 核蛋白40-60μg / 全细胞60-80μg

Something cool happening in Antibody Selection for Western Blot:

90% of Western Blot failures stem from poor antibody selection. After running hundreds of WB gels, here are 5 critical principles I've learned:

1️⃣ Match your application: WB requires antibodies that recognize linear epitopes under denaturing conditions — not all "IHC-validated" antibodies work for Western Blot.

2️⃣ Species compatibility: Never use mouse primary antibodies on mouse tissue samples without special protocols — secondary antibodies will bind endogenous IgGs, creating background chaos.

3️⃣ Monoclonal vs Polyclonal: Monoclonal = high specificity, batch consistency, ideal for quantitative work. Polyclonal = higher sensitivity but risk of cross-reactivity.

4️⃣ Optimize dilution: Higher antibody dilution reduces non-specific binding. Start at 2x the manufacturer's recommended concentration and titrate.

5️⃣ Always include controls: Every blot needs a positive control expressing your target protein.

The bottom line: Good antibodies are half the battle. Invest time in proper antibody validation — it saves more time than troubleshooting failed experiments.

Here are the top 5 Western Blot pitfalls I've learned to avoid after years of running gels:

🔬 Sample Prep: Keep RIPA on ice, lyse ≥30 min — don't skip this!
🫧 Transfer: Bubbles kill results. Roll firmly before transfer.
🎯 Antibodies: Titrate your primary Ab every time.
🛡️ Blocking: 1 hr BSA = sweet spot. More isn't better!
📸 Exposure: Capture a time series (10s-5min) for clean data.
