RDP3 is a peptide derived from rice with tissue-penetrating capability, which can promote repair of oral ulcer mucosal in vivo and in vitro. Mechanistically, RDP3 functions as an antagonist of the interleukin-2 receptor β subunit (IL-2Rβ) with a binding affinity KD of 0.99 μM. This leads to suppression of IL-2Rβ/PI3K activity and blocks pathological PI3K signaling, inhibiting NLRP3/GSDMD-mediated pyroptosis, reducing inflammation, and promoting mucosal regeneration. RDP3 can not only accelerate wound healing but can also restore microbiome homeostasis.
