Alzheimer’s disease is a debilitating condition that severely impacts not just the quantity but also the quality of life of patients and their families. Despite decades of research, most clinical trials of Alzheimer’s drugs, including anti-amyloid antibodies, failed to improve cognitive symptoms. However, recently Eisai and Biogen announced positive results from the phase 3 trial of lecanemab, a monoclonal antibody targeting amyloid-beta. On the face of it, it seems like Biogen and Eisai were finally able to develop the first truly disease-modifying therapy for Alzheimer’s. However, in reality, that may not be the full story. Is lecanemab really effective and what are the concerns around it? What makes it different from all other anti-amyloid drugs that failed in clinical trials?

Today most people have heard about gene editing, but have you heard about epigenome editing? Epigenome editing is a novel way of manipulating gene expression without permanently changing the DNA sequence. The potentially enhanced safety profile and versatility of the epigenome editing platforms have attracted the attention of scientists and investors alike, resulting in the emergence of four new epigenome editing companies, which since 2020 collectively raised $240 million from private and institutional investors. In this report, we highlight eight companies leading the innovation in epigenome editing including Chroma Medicine, Tune Therapeutics, Epic Bio, Sangamo Therapeutics, Omega Therapeutics, Navega Therapeutics and others and discuss how their platforms will lead to the rise of a new type of epigenetic medicines.

Our 45th politics update coincidentally reports on the official 2024 comeback attempt by the 45th President: we examine whether calls by senior Republicans for the party to move on from Donald Trump after the midterms misjudge the feelings of their membership. Would somebody like Ron DeSantis be more ‘unifying’ or do Trump’s base prefer party unity on their terms: one that displaces the McCarthy-McConnell ‘authority’ and completes the GOP’s populist shift at all levels? We finish with an outline of two problems facing the Republicans whoever their next presidential candidate is: a post-Roe polling deficit among young and female voters and a lag behind Democrats in adapting to the expansion of early voting.

This note is a summary of remarks given at a Primary event by the economist Prof. Patrick Minford; he gave his view on what the abandonment of the Truss/Kwarteng tax plans meant for the outlook of UK growth and argued they could have succeeded with better planning – but also with a Treasury more willing to cooperate. Downbeat on the fiscal agenda of Rishi Sunak and Jeremy Hunt, his discussion continued with examination of what ‘mandates’ meant when prime ministers could change without voting and the dynamic between elected officials and civil servants was so altered as of late.
