Forward vs Reverse Pipetting
Both are techniques used with micropipettes, but they differ in how much liquid is aspirated and dispensed.
🧪 Forward pipetting — steps
1. Press the plunger to the first stop.
2. Immerse the tip in the liquid.
3. Slowly release the plunger to aspirate the set volume.
4. Move to the receiving tube.
5. Press to the first stop, then second stop to completely dispense.
6. Withdraw the tip.
Example: Setting pipette to 100 µL → aspirates 100 µL → dispenses 100 µL.
🧪 Reverse pipetting — steps
1. Press the plunger to the second stop.
2. Immerse the tip in the liquid.
3. Slowly release the plunger to aspirate more than the set volume.
4. Dispense by pressing only to the first stop.
5. The excess liquid remaining in the tip is discarded.
Example: Setting to 100 µL → aspirates slightly more than 100 µL → dispenses 100 µL, with the excess left in the tip.
