TOMOCUBE See your samples like never before with Tomocube’s revolutionary label-free, live cell Holotomography

Established in 2016, Tomocube is dedicated to delivering products that enhance biological and medical research via novel optical solutions that can assist scientists and clinicians in understanding, diagnosing, and treating human diseases. Holotomography (HT) technology provides label-free 4D quantitative imaging for analyzing cells and tissues. The details of live cell dynamics, subcellular organelles, and tissue structures can be seen without preparation including fixation, transfection, or staining. Image data acquisition is supported by AI-driven data analysis software for phenotypic analysis using cellular and sub-cellular segmentation. How do you quantify lipid droplets inside a whole — without a single stain?

In a new study in Journal of Animal Science and Biotechnology (2026), researchers at Gansu Agricultural University used the HT-X1 to capture label-free 3D images of yak oocytes and measure lipid droplet volume directly.

Their finding: magnolol (MAG) increased that volume by 27% (P < 0.01) — part of the evidence behind how MAG improves yak oocyte maturation and blastocyst quality through a PPARγ-dependent pathway.

Most assessment still comes down to what a 2D image shows: how many cells there are and how even they look. A new study in Communications Biology asks what the inside of each cell can add.

Researchers from CHA Bundang Women's Medical Center, KAIST and imaged mouse embryos label-free on a customized HT-X1 every 12 hours, from the 2-cell stage to the expanded blastocyst, without affecting blastocyst rates in a standard mouse embryo assay.

Because (HT) measures refractive index (RI) in 3D, the team could quantify features that bright-field can't resolve:

🔹 Cytoplasmic RI heterogeneity, plus the size and spacing of high-RI foci, at 24 h

🔹 Individual nuclei in early morulae at 36 h, a stage where bright-field loses clarity

🔹 Dry mass density of each blastomere, calculated from RI

Embryos that later arrested had a more uneven cytoplasm and fewer, larger, more loosely packed nuclei. The team combined three HT-derived features with four morphology features obtainable by conventional imaging. With that combined set, a random-forest model predicted blastocyst formation with an AUC of 0.963, compared with 0.908 for morphology alone, in this mouse proof-of-concept.

We're bringing label-free 3D imaging to Seoul — CHA BIO Complex, Pangyo, Korea · Sep 17–18.

If you're working with organoids, stop by Exhibition Booth B2F #19 to see holotomography imaging and talk applications with our team.

🎤 Don't miss our talk:
Sep 18 (Day 2) · 14:05 · Dr. Sumin Lee, Senior Vice President
"Tomocube: Holotomography for label-free 3D quantitative organoid imaging"

Come see what living organoids look like in 3D — no stains, no fixing, no clearing. Track growth, drug response, and morphology over time, in the same living organoid, without disturbing it.

The organoid on our banner? A human small intestine organoid, captured label-free with its internal architecture fully intact.

We ran a workshop at the University of Alberta's Cross Cancer Institute this week with the HT-X1™ mini — the room was packed.

And what happens when you let the cells tell the story uninterrupted? Here are a few of the timelapses we captured:

🔹Glioblastoma tumor neurospheres, reaching toward one another and building tumor microtubes — the networks glioblastoma uses to share resources and resist treatment. Live and label-free, no stains.

🔹Huh-7 liver cancer cells captured using hetero-timelapse (holotomography with correlative fluorescence imaging at a lower frequency to minimize phototoxicity), showing lipid droplet formation under DGAT1 versus DGAT2 inhibition.

Both overnight, on living cells.

The workshop was a great success. Thanks to Dr. Xuejun Sun, Director of the Cell Imaging Facility, and Dr. Hilmar Strickfaden, Cell Imaging Core Director, for making time to stop by — and for letting us share these images. And a big thank-you to all our participants for bringing their samples and generously sharing this demo data with us.

𝗡𝘂𝗱𝗴𝗶𝗻𝗴 𝗮 𝘀𝗸𝗶𝗻 𝗰𝗲𝗹𝗹 𝘁𝗼𝘄𝗮𝗿𝗱 𝗮 𝗻𝗲𝘂𝗿𝗼𝗻𝗮𝗹 𝗳𝗮𝘁𝗲 — does it matter whose skin it came from?

Reprogramming skin fibroblasts into neurons could help repair damaged nervous tissue, but results vary by donor age and species — and the reasons are still not fully understood.

In a collaboration between CNR (Bologna) and the Barman Lab at Johns Hopkins, researchers grew mouse and human fibroblasts from young and elderly donors on two candidate materials: soft silk fibroin and stiff, rough hydrotalcite.

They used holotomography to measure how each surface reshaped the cells. The stiff hydrotalcite drove the strongest stretch toward a neuron-like shape. Human cells responded far more than mouse cells, and younger donors more than elderly — yet even elderly fibroblasts could still be nudged toward neuron-like shapes on hydrotalcite.

Holotomography provided the study's core morphological measurements — dry mass, cell area, and aspect ratio for every cell — turning a comparison that used to rest on visual impressions into something they could quantify.

🔬 Dose-response, treatment vs. control, or a multi-well screen.

Multi View in TomoAnalysis™ Viewer puts those conditions in front of you at once, navigation synced across every panel, so the answer shows itself in the data — not in a figure you assemble hours later.

🔹Line up a concentration series and see where RI distribution and morphology start to change.
🔹Compare structural changes at the same timepoint, side by side.
🔹Scan condition-dependent differences across wells in a single pass.
🔹Overlay fluorescence in each panel when it's available.

See the comparison directly — no switching tabs, no screenshots, no aligning images by hand.
