Publication highlight: Huber et al. Cancer Cell. 2024

This study from Ruben van Boxtel’s group, in collaboration with Hans Clevers, demonstrated improved detection of colibactin-induced mutations by genotoxic E. coli in organoids and colorectal cancer.

In this study, Huber et al., leveraged ResolveDNA® for single-cell whole genome sequencing of intestinal organoids exposed to different strains of E. coli.

Over 85% of CRISPR off-target indels detected using ResolveDNA® were missed by an in silico prediction method.

When it comes to CRISPR gene editing, the term "off-target edits" often enters the conversation. But what are they, and why do they matter?

Off-target edits refer to unintended DNA changes that occur at genomic locations other than the CRISPR target location. These edits can happen when the guide RNA directs Cas9 to a sequence with less-than-perfect homology, or even without any apparent sequence match. Factors like impurities in guide RNA preparations or previously unrecognized sequence differences between the genomes of the edited cells and the reference genome used during guide RNA design can also contribute to these off-target modifications.

Understanding off-target edits is critical because they can negatively impact interpretation of experimental results when using these cells in research and in the context of gene-edited medicines, off-target edits can reduce efficacy or pose serious safety risks.
