Neuroinflammation is a shared hallmark of Alzheimer's, Parkinson's, and multiple sclerosis, all of which involve blood–brain barrier breakdown that admits peripheral immune cells into the CNS. SRM University, AP, CSIR - Indian Institute of Chemical Biology, Jadavpur University, University of South Florida, and NEB researchers present an end-to-end framework to identify cell-specific protein markers commonly regulated across mild-moderate AD, early-stage PD, and MS in peripheral blood mononuclear cells. This framework selects candidates by shared differential expression, filters for BBB permeability, and maps them to neural cell types. Astrocyte- and monocyte-like PBMC populations showed transcriptional and functional convergence with macrophages and neuroglia, and pathway analysis flagged kinase–integrin interactions as key regulators. Seed markers, mainly kinases and immunoglobulins, were then assessed for druggable structural features and moonlighting behavior, giving a strategy for immune-based diagnostic and therapeutic target discovery.
