A conformational switch explains how small molecules can selectively inhibit TREX1. TREX1, the major cytosolic exonuclease, acts as a checkpoint for stimulation of the cGAS-STING pathway and is being targeted to enhance tumor immunity. Patricia C. Hernandez and colleagues at the University of Minnesota used X-ray crystallography to show that the inhibitors competitively block binding of the DNA substrate's 3′-terminal nucleotide. Inhibitor binding also stabilizes Tyr129 in an alternative conformation that is incompatible with DNA binding, creating a deep hydrophobic pocket that helps explain selectivity for TREX1 over TREX2 and related viral nucleases.
