An RNA helicase plays a key role in drug resistance in KRAS^G12C cancers: accumulated sotorasib-bound KRAS^G12C interacts with the RNA helicase, DHX9, trapping it in the cytoplasm and activating RAC1–PAK1 signaling. Importantly, inhibiting DHX9 helicase activity restored sotorasib sensitivity in resistant tumor cells.
