Volumetric time-course analysis of chemotherapeutic dose-response in 3D hydrogel embedded cancer spheroids using VitroPrime™ 3D Culture and Imaging Plates together with CYTOQUBE light-sheet microplate cytometer.

This work tracks oxidative stress, apoptosis, and necrosis in U87 3D cultures over 96 hours using an integrated workflow combining 𝗩𝗶𝘁𝗿𝗼𝗚𝗲𝗹® 𝗚𝗕𝗠 𝗫𝗲𝗻𝗼-𝗙𝗿𝗲𝗲 𝗘𝗠𝗧 𝗞𝗶𝘁, 𝗩𝗶𝘁𝗿𝗼𝗣𝗿𝗶𝗺𝗲™ 𝟯𝗗 𝗖𝘂𝗹𝘁𝘂𝗿𝗲 𝗮𝗻𝗱 𝗜𝗺𝗮𝗴𝗶𝗻𝗴 𝗣𝗹𝗮𝘁𝗲𝘀, and Hamamatsu's 𝗖𝗬𝗧𝗢𝗤𝗨𝗕𝗘 light-sheet technology, with dose-dependent cell death confirmed at TMZ concentrations known to be toxic to GBM cells.

Researchers at Chungnam National University investigated how the physical forces within a tumor, specifically mechanical stimulation, contribute to the aggressive invasion in glioblastoma (GBM). Using a combination of ATP imaging and electrophysiology, the team first established that mechanical stimulation causes GBM cells to release ATP.

To directly test the functional role of TRPM3 in invasion, the researchers performed invasion assays using the 𝘅𝗲𝗻𝗼-𝗳𝗿𝗲𝗲 𝗩𝗶𝘁𝗿𝗼𝗚𝗲𝗹® 𝗛𝘆𝗱𝗿𝗼𝗴𝗲𝗹 𝗠𝗮𝘁𝗿𝗶𝘅. They found that genetically silencing TRPM3 in U-87 MG GBM cells significantly reduced their ability to invade through the VitroGel® matrix. This critical experiment directly linked TRPM3 activity and subsequent ATP release to the invasive phenotype. This study concludes that the 𝗩𝗶𝘁𝗿𝗼𝗚𝗲𝗹® 𝗛𝘆𝗱𝗿𝗼𝗴𝗲𝗹 𝗠𝗮𝘁𝗿𝗶𝘅 was instrumental in validating the functional role of TRPM3 in GBM invasion. By 𝗽𝗿𝗼𝘃𝗶𝗱𝗶𝗻𝗴 𝗮 𝗰𝗼𝗻𝘀𝗶𝘀𝘁𝗲𝗻𝘁 𝗮𝗻𝗱 𝗲𝗮𝘀𝘆-𝘁𝗼-𝘂𝘀𝗲 𝟯𝗗 𝗰𝘂𝗹𝘁𝘂𝗿𝗲 𝘀𝘆𝘀𝘁𝗲𝗺, it enabled the researchers to clearly demonstrate that TRPM3-mediated ATP release is a key driver of invasion, positioning TRPM3 as a promising new therapeutic target.
