New study led by Dr. Denggang Fu and Dr. Sophie Paczesny at the Medical University of South Carolina (MUSC) reveals how leukemia stem cells (LSCs) drive both disease progression and resistance to treatment.

The research indicates that LSCs establish a self-sustaining loop through IL-33/IL-1RL1 signaling, enabling them to survive while shaping a suppressive immune environment in the bone marrow. This loop not only fuels leukemia growth but also shields it from the body’s defenses.

IL1RL1 emerges as a promising therapeutic target in acute myeloid leukemia (AML), with potential for next-generation bispecific antibody immunotherapies that can disrupt this vicious cycle.

In this study, the Monoclonal Antibody to Mouse T1/ST2, PE (Product Code: 101001PE) from MD Bioproducts was used to identify critical immune cell populations within the leukemic niche.

Can two treatments work better together to protect the kidneys in diabetes? That’s what a team of researchers from Université Bourgogne Europe and INSERM in Dijon, France, led by Océane Pointeau and Tony Jourdan, set out to explore.

They studied diabetic nephropathy (DN), a kidney disease caused by diabetes. The team tested a combination of two known therapies:

Empagliflozin, a medicine used to lower blood sugar
Monlunabant, a compound that blocks CB1 receptors, which are involved in inflammation and kidney damage

They treated diabetic mice with each compound alone or in combination. The combo therapy worked best, showing stronger protection against kidney damage, inflammation, and fibrosis (scarring of kidney tissue).

To support their findings, the team measured Uromodulin (UMOD, THP) levels in urine, a key biomarker of kidney health, using the MD Bioproducts Uromodulin ELISA kit (UMOD, THP).

This study demonstrates that targeting both blood sugar control and inflammation pathways may provide more effective protection for the kidneys in diabetic patients.

The MD Bioproducts Uromodulin (UMOD, THP) ELISA is a 96-well assay designed to accurately quantify Uromodulin levels in urine, plasma, and serum. This biomarker is essential for assessing kidney function, cardiovascular health, and metabolic disorders. The Uromodulin assay serves as a supporting tool for clinical studies and supports research related to acute kidney injury (AKI), chronic kidney disease (CKD), cardiovascular disease (CVD), and metabolic disorders.

A research team led by Chihiro Hara-Tsuchiya at Astellas Pharma Inc., Japan, has developed a novel screening system to identify compounds that regulate type 2 innate lymphoid cells (ILC2s) cells known to contribute to allergic diseases through the production of IL-4, IL-5, and IL-13.

Study findings:
- A high-throughput screening of ~30,000 compounds led to the discovery of AS4040605, a small molecule that inhibits both mouse and human ILC2 activity.
- The compound targets retinoic acid receptor-α (RARα) and may act as an inverse agonist.
- A robust IL-5 assay system was established using IL-2 + IL-33-stimulated mouse lung ILC2s.
- This approach offers a promising direction for developing future therapies against allergic diseases.

Throughout the study, mouse ILC2s were characterized using the Monoclonal Antibody to Mouse T1/ST2, FITC (Product code: 101001F) from MD Bioproducts.

This work highlights the value of functional screening systems in identifying novel immunoregulatory compounds and suggests that RARα inverse agonism is a viable therapeutic mechanism.
