This study identifies key regulators of IL-2Rα expression in human CD4+ T cells, revealing that their effects vary across T cell types and activation states. Using CRISPR screens and single-cell transcriptomics, the authors highlight MED12, a Mediator complex component, as a crucial regulator of gene networks in T cells. MED12 interacts with the COMPASS complex, influencing histone methylation and expression of key factors like KLF2 and MYC. 🧬
