AHA 2025 Recap: People are shutting off the water, but NO ONE is draining the sink.

The core data highlighted at the 2025 American Heart Association (AHA) Annual Meeting was focused on the positive outcomes from some of the LDL lowering therapies such as Merck’s Enlicitide. While these ARE positive data points, the unfortunate truth is NONE of these are tackling the root cause of cardiovascular related events, plaque buildup.

At Repair Biotechnologies, our focus is on PLAQUE REGRESSION and significant reduction of the plaque that, when ruptured, causes these significant cardiovascular related events, such as heart attack or stroke. And while these LDL lowering therapies do slow down the buildup of plaque, they have yet to meaningfully show plaque regression which means patients are still at risk if when their LDL is lowered (i.e. they are turning off the water, but not draining the sink).

IF YOU ARE INTERESTED to learn how Repair Biotechnologies is ‘draining the sink’, we invite you to not only visit our website, but also, sign up below to be added to our direct distribution where you can receive periodic updates as we continue to advance our lead candidates, REP-0003 and REP-0004, and as we aim to provide a TRULY CURATIVE SOLUTION for those at risk.

🧬 Breaking New Ground in Cardiovascular Therapy at AHA 2025 🧬

We’re thrilled to share that Repair Biotechnologies has presented compelling preclinical data at the American Heart Association (AHA) Scientific Sessions 2025, demonstrating that our lead candidate, REP‑0003, drives regression of established atherosclerotic plaque AFTER JUST SIX WEEKS of treatment, in mouse models.

🔍 Why does this matter?

--> REP-0003 is built on our proprietary Cholesterol Degrading Platform (CDP), designed to SAFELY break down excess intracellular free cholesterol, a key driver of plaque formation that conventional LDL-lowering therapies don’t target.

--> The effect seen in our study is rapid and plaque-regressing, marking a major leap toward treating high-unmet-need conditions such as homozygous familial hypercholesterolemia (HoFH).

--> Earlier this year, we were awarded Orphan Drug Designation (ODD) by the U.S. Food and Drug Administration (FDA) for REP-0003 in HoFH, underscoring the potential impact of this approach.

👏 Thank you to the AHA community for the platform to share our work and to our dedicated research team whose persistent efforts are translating bold science into meaningful therapeutic opportunities.

📌 What’s next?

We remain committed to advancing REP-0003 toward clinical development and to furthering our mission of treating disorders driven by toxic intracellular free cholesterol — from cardiovascular disease to metabolic dysfunction and beyond.

Did you know that one of the most persistent challenges in cardiovascular disease is the toxic cholesterol INSIDE your cells?

While today’s therapies focus on lowering LDL cholesterol in the bloodstream, they don’t address the root cause: excess free cholesterol trapped within cells that drives plaque growth, inflammation, and cardiovascular risk.

At Repair, our proprietary LNP-mRNA fusion protein therapy takes a fundamentally different approach:

🧬 Selective gene delivery targets affected tissues.
⚙️ Fusion proteins are produced inside cells to degrade toxic excess free cholesterol.
💥 Cholesterol breakdown and clearance restores cell health and reverses plaque formation.

In preclinical studies, this mechanism led to rapid plaque regression, improved cardiovascular performance, and reduced inflammation – results unmatched by conventional LDL-lowering therapies.

We’re not just slowing disease progression – we’re reversing it.

Have you heard about the latest in cardiovascular health?

Today’s standard of care in atherosclerosis is built around lowering risk factors: statins, PCSK9 inhibitors, ezetimibe, and others. These drugs can save lives, but they all share the same limitation: NONE address the root cause inside the artery wall.

At Repair Biotechnologies, we’re pioneering a different way to treat cardiovascular disease:

🔹 Not just lowering LDL - we target the toxic cholesterol inside cells that drives plaque growth and instability.
🔹 Complementary, not competitive - our therapy can work alongside statins or PCSK9 inhibitors to go further than risk management.
🔹 First true disease-modifying potential - aiming to regress existing plaque and stabilize arteries, not just slow new buildup.
🔹 Shifting the paradigm - from lifelong symptom control to the possibility of restoring vascular health.

Investor takeaway: We’re building what could be the first-in-class therapy to directly reverse atherosclerosis - tackling the biology that other therapies leave behind.

Don’t fall behind on the latest advancements, follow us today to learn more!

Repair Bio is Part of The Future of Cardiovascular Disease Treatment

ARK Invest just highlighted how curative therapies, like gene editing, could transform cardiovascular disease (CVD), moving beyond chronic therapies such as statins and PCSK9 inhibitors.

At Repair Biotechnologies, we share that vision. Our liver-directed platform is designed to:

Target root causes of disease biology, NOT just manage LDL-C levels.
Deliver durable, disease-modifying effects, NOT just another daily pill or injection.
Unlock a massive market opportunity, addressing the world’s #1 cause of death and $500B+ in U.S. annual healthcare costs.

What’s exciting is that while ARK points to gene editing, Repair’s approach could achieve similar “one-and-done” outcomes without gene editing. We’re pioneering a differentiated path to long-lasting impact for patients suffering from severe cardiovascular and metabolic disease.

For investors, the story is clear:
👉Repair Bio is aligned with the biggest healthcare trend of the decade, curative cardiovascular innovation, while taking a unique, highly differentiated route.

Read the ARK article here for context:

Cardiovascular disease is the world’s leading killer and one of the most economically damaging, as its prevalence and costs have been rising. While statins can reduce the risk of heart attacks by ~29%, residual risk and poor long-term adherence—roughly half of patients discontinuing after a year...

🧬 What is HoFH? and Why Should You Care?

HoFH is a rare genetic disease causing dangerously high LDL cholesterol from birth – often leading to HEART ATTACKS BEFORE AGE 20.

Current treatments fall short, and Repair Biotechnologies is advancing a therapy designed to functionally cure HoFH – not just manage it.

Our therapy could not only cure HoFH, but also, has the potential to help EVERYONE remove plaque build-up inside their arteries.

🚨 Repair Biotechnologies Secures Orphan Drug Designation 🚨

A major milestone — and a meaningful de-risking event for their lead asset.

The FDA's Orphan Drug Designation (ODD) isn’t just a regulatory win, it’s a commercial and strategic signal that the asset targets a serious unmet need with real potential.

For investors, here’s WHY this matters:
🔹 Regulatory Advantage – Accelerated pathways and agency support
🔹 Market Protection – 7 years of U.S. exclusivity post-approval
🔹 Economic Upside – Tax credits, waived fees, potential non-dilutive funding
🔹 Valuation Catalyst – Stronger positioning for partnering and Series B conversations

This designation enhances the company’s optionality — whether toward a faster clinical path, strategic partnering, or future capital raises.
