🔬 HPLC Method for Epibatidine Using Primesep 200
At SIELC Technologies, we have developed a reliable HPLC method for the analysis of epibatidine using the mixed-mode Primesep 200 stationary phase.

🔹 What is Epibatidine?
Epibatidine (C₁₁H₁₃ClN₃) is a highly potent alkaloid originally isolated from the Ecuadorian poison dart frog Ameerega anthonyi.
It exhibits strong nicotinic acetylcholine receptor activity and produces powerful analgesic effects. However, due to its extreme toxicity - potentially causing paralysis and respiratory arrest - it is not used therapeutically. Accurate chromatographic methods are essential for controlled research and analytical verification.

🔹 Why Primesep 200?
Primesep 200 is a mixed-mode analytical column with weak acidic ion-pairing groups embedded into long ligand chains.
✓ Enhances retention of strong basic compounds through cation-exchange mechanism
✓ Retains neutral compounds by reverse-phase mechanism
✓ Provides an additional interaction with polar molecules, enabling separation of isomers and structurally-related compounds
✓ Allows for mobile phase selection suitable for LC-MS and preparative chromatography, as well as low UV detection

⚙️ Typical HPLC Conditions
✅ Column: Primesep 200, 4.6 × 150 mm, 5 µm, 100 Å
✅ Mobile phase: MeCN/H₂O (30/70%)
✅ Modifier: 0.05% H₂SO₄
✅ Flow rate: 1.0 mL/min
✅ Detection: UV, 215 nm

🔬 HPLC Method for Analysis of Malathion Using Primesep SB
At SIELC Technologies, we’ve developed a reliable HPLC method for the analysis of malathion, a widely used organophosphate pesticide, using the mixed-mode Primesep SB stationary phase.

🔹 What is Malathion?
Malathion (C₁₀H₁₉O₆PS₂) is heavily used in agricultural and public health pest control, especially for mosquito and fruit fly management. Although considered relatively low in toxicity to humans, it metabolizes into malaoxon - a significantly more toxic compound - making accurate analytical monitoring important in environmental and food safety testing.

🔹 Why Use the Primesep SB Column?
Primesep SB is a reverse-phase analytical column with strong embedded basic ion-pairing groups. It contains a C18 carbon chain and basic quaternary amine groups.
✓ Enhances retention of acidic compounds through anion-exchange mechanism
✓ Separates hydrophobic basic molecules using a combination of ion-exclusion and reverse-phase mechanism
✓ Retains neutral compounds by reverse-phase mechanism.
✓ Offers mobile phases suitable for LC-MS and preparative chromatography, as well as low UV detection (working pH range from 1.0 to 4.5)

⚙️ Typical HPLC Method Parameters
✅ Column: Primesep SB, 4.6 × 150 mm, 5 µm, 100 Å
✅ Mobile Phase: MeCN/H₂O (40/60%)
✅ Buffer: H₂SO₄, - 0.1%
✅ Flow Rate: 1.0 mL/min
✅ Detection: UV, 194 nm
This method provides a straightforward and robust approach for pesticide monitoring in environmental, regulatory, and food safety laboratories.

🔬 High Performance Liquid Chromatography (HPLC) Method for Analysis of NAD and NADH
At SIELC Technologies, we’ve developed a simple and reliable HPLC method for the separation and UV detection of Nicotinamide Adenine Dinucleotide (NAD⁺) and its reduced form NADH using a PEI stationary phase.

🔹 What are NAD and NADH?
Nicotinamide adenine dinucleotide (NAD) is an essential coenzyme present in all living cells. It exists in two interconvertible forms:
– NAD⁺ (oxidized form)
– NADH (reduced form)
The reversible conversion between NAD⁺ and NADH plays a central role in cellular redox reactions, energy metabolism, and electron transfer processes.

🔹 Why Use the PEI Column?
SIELC has engineered a PEI-specific column that combines ion-exclusion and size-exclusion mechanisms. This unique selectivity enables:
✓ Effective retention and separation of NAD and NADH
✓ High selectivity toward highly polar, charged biomolecules
✓ Reliable separation from excess inorganic ions and matrix components
✓ Simple mobile phase composition with UV detection

⚙️ Typical HPLC Method Parameters
✅ Column: PEI, 4.6 × 50 mm, 5 µm, 100 Å
✅ Mobile Phase: – MeCN/H2O – 60/40%
✅ Buffer: H₂SO₄, 0.2%
✅ Flow Rate: 1.0 mL/min
✅ Detection: UV, 264 nm

This method provides a straightforward and robust approach for NAD/NADH analysis in biochemical, pharmaceutical, and life science applications.

HPLC Method for Phospholipids on Lipak™
Phospholipid analysis is never trivial. Structural diversity, amphiphilic behavior, and complex matrices often mean long method development and constant optimization.

Lipak™ was designed to change that.
Lipak™ employs mixed-mode chromatography, combining reversed-phase interactions with a highly polar functional group in the stationary phase. This dual-mode interaction enables efficient separation of lipids and other highly hydrophobic compounds, even within complex mixtures.

Why Lipak™
• Retention of both nonpolar and amphipathic lipid species
• High resolution with excellent reproducibility
• Improved selectivity across diverse lipid classes
• Optimized performance with high-organic mobile phases
• Inert hardware coating to reduce peak tailing
• Available in dual-ended and new single-ended designs

🔬 High Performance Liquid Chromatography (HPLC-MS) Method for Analysis of Sodium Dodecyl Sulfate (SDS)

At SIELC Technologies, we’ve developed a robust HPLC-MS method for the analysis of Sodium dodecyl sulfate (SDS) using the Lipak stationary phase - an effective solution for surfactant analysis in complex matrices.

🔹 What is Sodium Dodecyl Sulfate (SDS)?

– Also known as Sodium Lauryl Sulfate (SLS)

– Molecular formula: C₁₂H₂₅NaSO₄

– Structure: An anionic surfactant with a hydrophobic C12 alkyl chain and a polar sulfate group

– Applications:

• Cleaning and personal care products

• Food industry (emulsifier and whipping aid)

• Pharmaceuticals (ionic solubilizer and emulsifier)

• Laboratory workflows (cell lysis for RNA/DNA extraction)

🔹 Why Use the Lipak Column?

The Lipak column employs mixed-mode chromatography, combining reverse-phase interactions with a negatively charged group in the stationary phase. This enables:

✓ Simultaneous retention of polar and nonpolar species

✓ High resolution and reproducibility for complex lipid and surfactant mixtures

✓ LC-MS compatibility for confident identification and quantification

⚙️ Typical HPLC-MS Method Parameters

✅ Column: Lipak, 3.2 × 150 mm, 3 µm, 100 Å, dual-ended

✅ Mobile Phase: MeCN/H₂O gradient (60/40 → 90/10)

✅ Buffer: Ammonium formate 10 mM + 0.05% formic acid

✅ Flow Rate: 0.5 mL/min

✅ Detection: LC-MS, ESI, SIM, m/z 265 [M–H]⁻

✅ LOD: 8 ppm

*LOD was determined for this specific instrument, method, and analyte; values may vary between laboratories.
