Did you know June 16th is National Fresh Veggies Day? This application note highlights the analysis of spinach which is one of the most nutrient-dense leafy greens available globally.

Characterizing fat-soluble vitamins in complex botanical matrices requires high-resolution stationary phases. In this application, a spinach sample was extracted in hexane and analyzed using a HALO® C30 column. To demonstrate baseline resolution under efficient isocratic HPLC conditions, the matrix was evaluated alongside a standard spike of Vitamins A, D2, and K1.

The shape-selectivity of the HALO® C30 phase provides the critical resolution needed for complex hydrophobic compounds, making it an excellent choice for food quality and nutritional analysis.

In recognition of National Candy Month, our latest application note focuses on sugar profile analysis in complex food matrices. Quantifying sucrose and lactose across different chocolate formulations requires a highly selective stationary phase capable of handling complex fat and protein matrices.

We demonstrate the separation of these key carbohydrates in milk, white, and dark chocolate using HILIC mode coupled with an evaporative light scattering detector (ELSD). The workflow utilizes the HALO® Penta-HILIC column, which features a polar ligand with five hydroxyl groups tethered to Fused-Core® silica particles for enhanced selectivity of hydrophilic compounds.

While most LCMS methods are performed in positive polarity, some such as oligonucleotide analysis are typically run at high pH in negative mode. This can invite the formation of metal adduction from the presence of Na+, K+, and others which can complicate the interpretation of MS data.

There are a number of steps you can take to minimize the presence of these cations such as the use of Type-1, 18 MOhm water, minimizing the use of borosilicate glass for storage of buffers, using polycarbonate instead, and using LCMS grade reagents that are certified free of alkali metals.

For example, hexafluoroisopropanol, which is commonly used in oligonucleotide LCMS buffers can frequently contain considerable amounts of alkali. Using LCMS grade such as the Supelco Lichropur HFIP, will make your results much easier to interpret and reliable.
