Biortus is excited to announce the successful determination of the high-resolution crystal structure of TNIK at 2.70 Å.

TNIK is a key regulator of the Wnt/β-catenin and Hippo signaling pathways and has emerged as a promising therapeutic target in oncology and neurodegenerative disease research. The availability of this high-quality structure provides valuable insights for structure-based drug design (SBDD) and target-focused drug discovery. Based on the email draft, the structure was determined by X-ray crystallography at 2.70 Å resolution.

Biortus Structural Biology Library New Releases
Protain: FASN| Resolution: 2.18 Å | Method: X-ray Diffraction

Biortus proudly announces the high-resolution crystal structure of FASN (Fatty Acid Synthase), determined at 2.18 Å by X-ray crystallography.

Fatty Acid Synthase (FASN), a key lipid-metabolizing enzyme, catalyzes the production of saturated fatty acids from acetyl-CoA and malonyl-CoA. It modulates the survival, differentiation, and function of multiple immune cells, thereby playing a role in the pathogenesis of immune-related diseases. Aberrant FASN expression and its synthesized fatty acids are observed in tumors, cardiovascular disorders, and inflammatory and autoimmune diseases.

Biortus has successfully determined the high-resolution apo FASN structure, providing key structural information to guide structure-based drug design and facilitating the rational development and optimization of FASN-targeted small-molecule inhibitors.

High-Resolution Cryo-EM Structures Reveal the Assembly Pathway of HSP60

How does HSP60 assemble into its functional double-ring chaperonin complex?

Capturing the transient intermediates that form during this process has remained a major challenge in structural biology. Our team at Biortus has successfully resolved the cryo-EM structures of three closed-ring HSP60 oligomeric states:
🔹 Hexamer (2.83 Å)
🔹 Heptamer (3.22 Å)
🔹 Tetradecamer double-ring assembly (2.61 Å)

Notably, clear ATP density was observed in all three structures, providing valuable insights into the molecular mechanism of HSP60 assembly and function.

This work highlights Biortus' capability in resolving complex macromolecular assemblies and multiple conformational states through high-resolution Cryo-EM.

Biortus is proud to announce the successful determination of the human adenosine receptor A3AR in complex with the ligand CF101 in its active conformation at an outstanding 2.81 Å resolution using cryo-electron microscopy (Cryo-EM).

The A3AR–CD39 signaling axis plays a critical role in brain repair associated with vascular dementia (VaD). CF101 (Piclidenoson)—a selective A3AR agonist currently in Phase III clinical trials for psoriasis—has demonstrated the ability to promote brain tissue regeneration and improve memory and motor function, pointing to a promising therapeutic strategy for VaD.

The A3AR–CF101 cryo-EM structure solved by Biortus (EMD-66627, 2.81 Å) is the highest-resolution structure available in this receptor family within the EMDB. The map resolves side-chain interactions within the A3AR binding pocket with exceptional clarity, revealing how CF101 engages and activates the receptor—providing an important structural framework for understanding CF101-mediated signaling and receptor activation.

This achievement further strengthens Biortus’s position as a global leader in structural biology and drug discovery. As the world’s largest commercial Cryo-EM facility, Biortus is equipped with multiple Titan Krios G4 instruments, supported by Glacios, Talos, and TF20 platforms, and a team of expert scientists experienced in solving atomic-resolution structures of large and complex biomolecules.

We’re excited to share the high-resolution crystal structure of LANCL2 (Lanthionine Synthetase C-Like Protein 2), solved at 2.15 Å by X-ray crystallography.

🧬 Why LANCL2 matters
LANCL2 is an abscisic acid (ABA) receptor expressed in immune, epithelial, and muscle cells. It regulates insulin sensitivity, glucose metabolism, and immune responses through NF-κB inhibition.
Its dysregulation has been linked to metabolic disorders, inflammatory diseases, and cancer — making this structure a valuable foundation for LANCL2-targeted drug discovery.
