Extracellular vesicles (EVs), including exosomes and microvesicles, are nanosized structures released in the extracellular space. EVs are released by many cell types, are present in all body fluids, and are involved in intercellular communications. EVs embody the status of the producer cells. Thus, they may serve as i) indicators of health or disease states, ii) minimally invasive biomarkers for disease diagnosis, or iii) predictors of therapeutic response. Despite the importance of EVs, current EV isolation methods have limitations. Dr. Okeoma’s webinar will describe how her lab developed particle purification liquid chromatography (PPLC) as a superior EV isolation tool and applied it to discover that circulating blood plasma-derived EVs (BEVs) hold promise as minimally invasive biomarkers for predicting response to neoadjuvant chemotherapy in patients with breast cancer. Her findings have direct relevance to clinical management of breast cancer and likely other malignancies.

RAS genes have gained prominence due to their implication in human cancer. Mammals harbor three RAS loci (HRAS, NRAS and KRAS) that are found mutated in many different tumor types, albeit with clear tissue-selective preferences for individual RAS loci. Moreover, whereas HRAS and NRAS produce only one major protein, the KRAS locus gives rise to two different protein isoforms via alternative splicing of the final exon. Thus, all four RAS proteins differ in the C-terminal hypervariable region that determines how RAS proteins are anchored to the cell membrane. Despite these differences, RAS proteins share common biochemical features and participate in similar activities. In his webinar, Dr. Drosten will focus on three fundamental aspects of RAS biology: (1) What is the biological difference between the three RAS genes and what is their role in normal homeostasis? (2) What is the (differential) role of RAS paralogs in cancer? (3) What are the contributions of each of the KRAS isoforms to development and cancer? He will explain how mouse models have been crucial in revising some of the classical concepts of RAS biology.

The extraordinary clinical potential of regenerative medicine will only be fully realized once there is a clear understanding of how to reliably manipulate stem cells. Accomplishing this will require a mechanistic appreciation of the interplay between stem cells and their respective natural niches. Dr. Matthew Tierney has studied stem cell-niche dynamics in different physiological and anatomical settings throughout his young career. Beginning with research in exercise physiology, he focused on understanding the cues that define the interactions between stem cells and the skeletal muscle microenvironment. From there he moved on to the hair follicle, where he is presently studying the signals that mediate stem cell self-renewal, differentiation, and wound-related plasticity. In his webinar, he will describe his identification of niche signals that lie at the crossroads of homeostatic regeneration and wound-enabled plasticity in the skin, thereby establishing a functional model for stem cell-niche crosstalk investigation.

Technological advances in the field of RNA and RNA-binding protein (RBP) biology are revealing not only the vast number of RBPs, but also the diverse functions of RNA and RBPs in normal physiology and disease. Dr. Mathias Munschauer began studying RNA and RNA-binding proteins while an undergraduate student and over the course of his subsequent training he helped to develop crucial techniques that have facilitated interrogation of the mRNA-bound proteome. Now in an independent position at the Helmholtz Institute for RNA-based Infection Research (HIRI) in Germany, he investigates RNA-protein at the host-pathogen interface with the goal of mechanistically defining RNA function during infection. In 2020, his group published a landmark paper detailing the SARS-CoV-2 RNA-protein interactome in Nature Microbiology, and that work will be the framework for his webinar.

GeneTex is excited to co-host this 2022 POSTDOC WEBINAR SERIES with LubioScience.

Cutaneous malignant melanoma is the most aggressive type of skin cancer and is responsible for approximately 75% of all skin tumor deaths. Inhibitors of the MAPK signaling pathway (MAPKi) have become important therapies to combat this disease, though resistance to these agents limits their long-term benefit. Understanding how MAPKi resistance arises will offer insight into how to defeat it.

Dr. Lorenza Ferretti’s work focuses on the discovery and characterization of epigenetic alterations responsible for MAPKi resistance. In this webinar, Dr. Ferretti will present her 3D drug screening approach in patient-derived melanoma cells and her compelling findings that PARP inhibition dramatically enhances the efficacy of MAPK inhibitors in cells previously resistant to these agents.

Inflammatory training is the generation of an epigenetic memory that leads to an augmented state of responsiveness to a broad range of secondary stimuli. Though previously thought to be exclusive to the innate immune system, Dr. Samantha Larsen discovered that epithelial stem cells can undergo inflammatory training that accelerates tissue repair. She revealed this phenomenon hinges on the coordinated efforts of stimulus-specific, stress-responsive, and homeostatic transcription factors that extend to diverse stimuli, cell types, and species. She has now extended her interests in the long-term consequences of inflammation to the brain, where she has identified that neurons, much like other long-lived cell types, undergo inflammatory training that may predispose them to subsequent neurodegeneration. Moving forward, she is addressing the persistent transcriptional and electrophysiological changes that follow concussive brain injury with the ultimate goal of treating and preventing injury-induced neuropathogenesis.

GeneTex is honored to host Dr. Bryan Ngo's presentation on Genetic and Metabolic Determinants of Tumor Evolution and Metastasis. Dr. Ngo’s research focuses on dissecting the complex interplay between genomic instability, nutrient constraints, and metabolic synthetic processes that impact the ability of a cancer cell to survive and proliferate in diverse metastatic tissue environments. In this webinar, he will describe two independent studies that (1) connect a form of genetic instability, namely chromosomal instability (CIN), with its causal role in inflammation-driven metastasis, and (2) uncover a novel therapeutic paradigm that exploits the interplay between extracellular nutrient availability and cell-intrinsic glucose-derived serine synthesis in the context of brain metastasis. Dr. Ngo’s work is expanding our knowledge of how chromosomal instability, innate immune signaling, and cancer metabolism drive metastasis, and how this insight can potentially offer new therapeutic approaches to treat metastatic disease.
