New publication: Targeting nucleolin makes glioblastoma cells fragile

Glioblastoma is among the most treatment-resistant brain cancers. In part because it can survive treatments like chemotherapy and radiation by rewiring its cells' metabolism. A new study targeted nucleolin, a protein overexpressed on glioblastoma cell surfaces, using AS1411. AS1411 is a short DNA aptamer, a folded strand of DNA engineered to bind a target with antibody-like precision.
This treatment shifted tumor cell membranes toward a more rigid, saturated lipid composition, while also enriching lipids that fuel ferroptosis, a form of cell death driven by lipid oxidation. Together, these changes create a potential opening: cells under this stress may become more susceptible to therapies that push them toward ferroptotic death.
