New research from Ben-Shalom, Freund and colleagues reveals the discovery of antibodies that can disrupt pro-survival signaling in melanoma cells, exposing a new therapeutic vulnerability.

Melanoma cells release melanosomes, specialized extracellular vesicles that normally transport melanin, but can be repurposed by cancer cells to deliver pro-tumorigenic signals to surrounding cells in the tumor microenvironment (i.e. a paracrine mechanism). The authors in this study demonstrated for the first time that melanosomes ๐˜ข๐˜ญ๐˜ด๐˜ฐ support the growth and survival of the same melanoma cells that produce them (i.e. an autocrine mechanism).

๐—ž๐—ฒ๐˜† ๐—ณ๐—ถ๐—ป๐—ฑ๐—ถ๐—ป๐—ด๐˜€:
โ€ข Immunization with melanosomes generated antibodies against extracellular HSP70, located on the melanosome vesicle surface. These antibodies acted similarly to neutralizing antibodies targeting viral antigens, blocking HSP70-mediated signaling and reducing melanoma cell survival.
โ€ข These anti-HSP70 antibodies inhibited melanoma growth ๐˜ช๐˜ฏ๐˜ฅ๐˜ฆ๐˜ฑ๐˜ฆ๐˜ฏ๐˜ฅ๐˜ฆ๐˜ฏ๐˜ต๐˜ญ๐˜บ of additional immune effector cells (like NK cells).
โ€ข Anti-HSP70 antibodies inhibited melanoma growth in mouse models, leading to delayed tumor development and improved survival.
โ€ข In metastatic melanoma patients receiving immune checkpoint therapy, stronger anti-HSP70 antibody responses were associated with complete responses, highlighting a potential link between antibody immunity and treatment outcomes.

This work positions melanosomes as a promising platform for therapeutic discovery and identifies extracellular HSP70 as a potential target for antibody-based melanoma therapies.
