🔹 One of the biggest misconceptions in cell therapy development is that a high post-thaw viability is an indication of successful cryopreservation. When we have discussed with many groups about cryopreservation optimization, a common response we hear is, “Our post-thaw viability is +90%, we are OK.”.

🔹 Delayed Onset Cell Death (DOCD) occurs when accumulated stressors during processing tip the cells over the tolerable threshold into activation of irreversible apoptosis. We have seen it all - with T cells, NK cells, iPSCs, and many more.

🔹 These stressors can start accumulating throughout the process starting from days approaching harvest, to freezing, and post-thaw steps.

In a new Pharmaceutical Executive Magazine article, BioLife’s Chief Technology Officer, Dr. Sean Werner, states that many promising cell and gene therapies do not fail because of safety or clinical efficacy problems, but because of manufacturing, quality-control, and regulatory issues that emerge late in development.

Dr. Werner explores how CGT developers often focus heavily on clinical success, while underestimating the importance of Chemistry, Manufacturing, and Controls (CMC), assay validation, and process consistency. By the time these issues surface during an FDA review, fixing them can be costly and time-consuming. 🕜
